AbstractMultiplex Polymerase Chain Reaction can playa major role in detection of seafood borne pathogens. This study describes the development of MPCR . system for the stimultaneous detection of Salmonella enteric a, Listeria monocygenes and Vibrio parahaemolyticus that are found in seafood. MPCR amplification of inv A gene of Salmonella enterica, hly gene of Listeria monocygenes, gyrB genes for Vibrio parahaemolyticus were used for detection. Four DNA extraction methods were studied and Triton X 100 method was found to be the best for all three indicator microorganisms. For simultaneous detection of these bacterial a annealing temperature of 58°C, DNA primer of concentration 2.5 IlM and volume of 2.5 III gave the best result on optimization. The sensitivity detection show that the lower detection limits were 104 cells / ml was obtained; enrichment with TSBYE broth for 6 hours, the detection limit 100 cells/ml. Thus, the MPCR system developed would help to ensure rapid and accurate detection of Salmonella enterica, Listeria monocygenes and Vibrio parahaemolyticus
