AbstractDuring the production of bio-diesel from vegetables oils, considerable amounts of glycerol are produced as a by-product. This is a major environmental concern. In this study the possibility of using glycerol for pre-treatment of rice straw before hydrolysis and fermentation to ethanol was studied. Optimization for the production of glucose from glycerol-pretreated rice straw was carried out. It was found out by using experimental design. The optimum condition for 24 hr and 48 hr pretreatment is found at pretreatment of 220 ⁰ C for 1 hr. Rice straw cut to size 1 cm and milled 20 mesh gave nearly the same reducing sugar yield. It means the size of rice straw between 1 cm and 20 mesh can not affect on the yield of reducing sugar. The short time pretreatment can give higher yield than the longer pretreatment time. Enzymatic hydrolysis yield not only depends on the pretreatment time but also on the temperature of glycerol pretreatment. Acid hydrolysis of pretreated rice straw give higher yield than enzymatic hydrolysis and the yield of it is 588 mg/ g dry weight of rice straw. The acid hydrolysis product was fermented by using Saccharomyces Cerevisae. The yield of glucose to ethanol is 0.31, so the stoichiometric yield from glucose is 61 % of theoretical yield. The glycerol pretreatment method for rice straw should be used as pretreatment method because it can make rice straw into thin and fine fibrils and it is accessible for hydrolysis. Hence glycerol, a by-product of biodiesel production can be used for pre-treatment of lignoellulosic residues
