AbstractIn this study, ultrasound - assisted extraction and microwave - assisted extraction were used for leaching the bioactive compounds from dried nettle leaves. The maximum phenolic content obtained from ultrasonic - assisted extraction was 33.43 mg GAE/g DW . Microwave - assisted extraction was found to be more effective than other conventional methods for the extraction of polyphenols from nettle leaves. 61.9mg GAE/g DW of polyphenol was extracted in 10min of microwave extraction using power supply of 100W. For the protein yield from alkaline ultrasound extraction, the maximum yield was 35.81 mg/g DW at 60°C, 30 min and 1.5 M of NaOH. All the extraction methods were optimized. The proximate, qualitative and quantitative phytochemical analysis of the dried nettle leaves were carried out. The presence of phytochemicals helped to understand the nutritional and pharmacological significance of the nettle plant. The nettle extracts were encapsulated using the calcium - alginate - chitosan system to investigate the influence on encapsulated bioactive extracts and their controlled release in simulated gastrointestinal system. The stability and encapsulation efficiency was found to be higher of about 75% in the multilayered beads, externally coated by chitosan. However, in the simulated gastric fluid (SGF, pH 1.2), the lower release of polyphenols was observed in both the single - layered calcium - alginate system and the double - coated with chitosan beads
