Abstract | Popular consumption of caffeine containing beverages in daily life not only adversely affects human health, but also may contaminate surrounding water. Decaffeination has been on demand these days in order to cut down caffeine contamination and to produce caffeine free products, especially coffee and tea beverages since ―coffee drinking behavior and habit‖ survey showed that 87% of residents of AIT campus like to drink coffee in their daily life. However, since conventional decaffeinating methods are expensive and sometimes involving toxic chemical agents, microbial caffeine degradation method has been emphasized and developed. Hence, caffeine degrading microbes take part in important role for bio-decaffeination. In this study, caffeine tolerant bacteria were isolated from the soil of coffee growing area. According to 16SrDNA analysis, it was identified as Burkholderia anthinawith the highest similarity 99.93% at nucleotide sequences among Burkholderia speices. It can tolerate high level of caffeine up to 20g/land can degrade 44% and 38% of caffeine content in media containing 5 g/land 10g/lof caffeine, and 14% and 10% at 15g/l and 20 g/lof caffeine containing media during 120 hours. Thus, Burkholderia anthina can be effectively used to degrade caffeine from agro-industrial wastes. |
