Qualitative analysis of tetracycline residues and evaluation of antibiogram of salmonella and vibrio isolates from whiteleg shrimp

AbstractSeafood is prone to bacterial contamination and could have some health hazards on consumers. The present thesis was aimed to study the occurrence of contaminated Salmonella and Vibrio in white - leg shrimp from Thala Thai Market (Pathumhtani Province). The study was carried out on 20 shrimp samples for isolati on and identification of Salmonella and Vibrio ; samples were c ultured on Xylose lysine desoxycholate (XLD) agar plates and Thiosulphate Citrate Bile Salt Sucrose (TCBS) agar was used for Vibrio isolation respectively. Four Salmonellae and three Vibrios wer e isolated out of twenty tested samples (20% and 15%) respectively. For knowing the antibiotic sensitivity (Antibiogram) of the isolates, the Kir - Baeur disk diffusion test was performed to determine the zone of inhibition. The resistance to tetracycline (TE T), Amoxicillin (AMX) and Ampicillin (AMP) was found in all four Salmonella isolates (100.0%), the resistance to sulfamethoxazole in two out of four Salmonella isolates (50.0%) and only the intermediate resistance to sulfamethoxazole was detected in all th ree Vibrio isolates (100.0%). Detection of some antimicrobial resistance genes was done by multiplex PCR and tetA genes responsible for tetracycline resistance were shown positive in 50.0% of all four Salmonella isolates but no tetB , blaCMY and blaSHV gene s were shown. Qualitative analysis of tetracycline residues was shown positive in five out of twenty shrimp samples (25.0%).
