The growth of freshwater microalgae for the production of polyunsaturated fatty acids

AbstractThis study had two major phases of work to determine the extent of effect of light and temperature for better growth performance and the development of some important polyunsaturated fatty acids (PUF A) in the cells of the freshwater microalgae. Growth studies (the first phase of the experiment) of four species of freshwater algae, Chlorella vulgaris, Scenedesmus obliquus, Synechocystis aquatilis and Chroococcus turgidus, were conducted in five 1-liter flasks culture set-ups. The species were grown in two conditions of light, 3 k lux and 6 klux, two temperature conditions, 24-26°C, and 32-36°C and under natural ambient condition, were BG-11 was used as media and nutrient source to all species grown. The effects on the growth was dete1mined by biomass monitoring in terms of ash free dry weight per ml of sample and the actual cell density in number of cells per ml sample. Results showed that all species have grown successfully for the duration of 8 days culture. The C. vulgaris, S. obliquus and S. aquatilis were found to obtain the highest cell mass and density at 6 k lux and 24-36°C or classified in this study as high-light low temperature condition, while Chroococcus turgidus grew best in 6 k lux, 34-36°C or low-light high temperature condition. The second phase was the analysis of samples for detection of fatty acid composition and concentration of the cells in all conditions. It was dete1mined that direct transterification procedure has given the most number of peaks as shown from the GC chromatogram. A total of 22 different fatty acids were identified. The predominant PUF A in all species is a-linolenic acid or C18:3m3. The two green algae, Chlorella vulgaris and Scenedesmus obliquus were found to obtain the highest yield equivalent at 3 klux, 34-65°C and 6 k lux at 24-26°C, respectively. Likewise, among the best growing conditions that enhanced the C18:3ro3 production, Chlorella vulgaris and Scenedesmus obliquus consistently got the highest concentration and yields of these important nutrients.
