AbstractThis research aimed to develop a multiplex PCR amplification of four species of foodbome pathogens for the individual and simultaneous detection both in pure culture and seafood samples. The four species of foodbome pathogens examined were E.coli 0157:H7, Listeria monocytogenes, Vibrio parahaemolyticus and Salmonella typhimorium. Simultaneous amplification of products of 398-bp, 482-bp, 622-bp and 685-bp length was obtained either by using mixtures of individual strains of E.coli 0157:H7, Listeria monocytogenes, Vibrio parahaemolyticus and Salmonella typhimorium or minced contaminated mussel/shrimp with all four bacteria. By using appropriate primers and optimized PCR conditions. The PCR protocol used universal culture medium and the same PCR conditions with a set of four pair specific designed primers. No interferences or non-specific amplification was observed using the PCR protocol when seafood samples were artificially inoculated with each individual or simultaneous bacteria species. The sensitivity and specificity of this protocol was evaluated. Multiplex PCR amplification was shown to be an effective, sensitive and rapid method for the simultaneous detection of foodbome pathogens in mussel and shrimp samples.
