Abstract | Lectin was isolated from the Abrus precatorius seeds by Ammonium sulfate precipitated, dialysis and Sepharose-4B affinity chromatography. The homogeneity of the lectin was demonstrated by SDS-PAGE in the presence of P-mercaptoethanol. Only one protein band with an apparent molecular mass of 60 kDa was found. The Abrus precatorius lectin showed carbohydrate specificity for D-galactose, a requirement for divalent metal rations (Ca2+ and Mn2+) for full activity. The hemagglutination activity was found to he stable up to 85°C and more stable under alkali than acid condition. Ouchterlony lmutnunodiffusion assays using human antiserum raised against the purified lectin of Abrus precatorius showed that it was possible to detect only IgA 1. The purified lectin was carried out to identify coaglulase-negative and coaglulasepositive staphylococci, Bacillus thuringiensis, Bacillus antluacis and Listeria monocytogenes serotype 3a. IgAl concentration was found to be higher in patients with liver cancer and hepatitis B compare to a value of 2.84 mg/ml in healthy person by using Enzyme - Linked -- Lectinosorbent - Assay (ELLA) method. Abrus precatorius lectin thus has a specific reaction with antibody IgAl and can be used for diagnosis |
