AbstractThe source of chitin deacetylase used in this study was Absidia coerulea (IFO 5301). The parameters studied in this work were the cultivation conditions for increasing enzyme production. Size and age of inoculum, time of incubation, composition of culture medium and pH effect on production of chitin deacetylase were investigated in shake flask fermentations. In the present study the sample which had 58.8% DD chitosan (1.5%) gave the highest activity as compared to glucose and N-acetyl D-glucosamine. However, the method of preparation of the chitosan sample is an important criteria for enzyme activity. Cultivation in glucose 2% medium and N-acetyl D-glucosamine 2% medium with initial pH 4.5 was optimal. The effect of peptone on enzyme production medium was also studied. The optimum inoculum age and inoculum size were found to be 5-day and 1.32 x 107 spores/ml respectively. The total activity of chitin deacetylase in culture filtrate of Absidia coerulea was maximal after 24 h in glucose 2% medium, after 32 h in 2% N-acetyl D-glucosamine medium, and 72 h in 1.5% chitosan 58.8% DD medium respectively. The total activity obtained from reactor study was nearly same value as shake flask fermentation due to wall growth and inadequate mixing condition. The morphology of the organism, which affects fermentation, is dependent on a number of parameters like nutrient source, pH, temperature, etc.
