AbstractImmobilization of concentrated glucosyltransferase enzyme on sodium alginate and copolymer of alginate/chitosan complex were studied. About 15-20% and 30% of immobilization efficiency were observed from immobilization by sodium alginate and alginate/chitosan complex, respectively. Suitable conditions for preparation of calcium alginate beads were 3% sodium alginate, 0.2 M CaC12 and incubation time of 1 h with 3 mm diameter of beads. Immobilized glucosyltransferase expressed better characteristics (i.e. pH stability or thermal stability) than free enzyme especially in the case of calcium alginate beads. Calcium alginate beads had greater stability than chitosan-coated calcium alginate beads and higher percentage panose production efficiency. However, chitosan-coated calcium alginate beads had higher storage stability and presented more enzyme activity than calcium alginate beads when the same amount of enzyme was immobilized. In batch reactor, 45.96% and 33.33 % panose production efficiency were obtained by calcium alginate beads and chitosan-coated calcium alginate beads respectively with 5% maltose as substrate.
