Abstract | A sensitive, simple, reliable and faster extraction method could be developed for the determination of anatoxin-a in the algae suspension water and freeze-dried cell. The excellent linearity with the correlation coefficient over 0.99 with both mobile phase solutions could be drawn using standard anatoxin-a. With 70%methanol+30%0.01M HCIO4 mobile phase solution and 1.2 ml/min flow rate, HPLC could detect the least toxic concentration of 0.34 mg/l (0.0034 µg/10µ1) and with methanol-water (6:4) mobile phase solution and 1 ml/min flow rate, HPLC could detect the least toxic concentration of 0.33 mg/l (0.0033 µg/10 µl). Combined with the extraction procedure which affected a 10-fold concentration and an overall assay sensitivity of 0.03 mg/l (0.3 ng/10µl) was achieved. HPLC with methanol-water (6:4) mobile phase solution, 1 ml/min flow rate could detect the highest amount of anatoxin-a combined with the developed extraction method (M.AA method). Anatoxin-a could be detected from the three species of the cyanobacteria namely Oscillatoria obscura TISTR 8245, Cylindrospermum TISTR 8158 and Cylindrospermum TISTR 8216 as a new investigation. |
