Abstract | Information on Salmonella contamination during production of shrimp, postharvest and handling on processed was collected. Relative performance of three Salmonella detection methods viz. the conventional method (AOAC), Modified Semi-solid Rappaport Vassiliadis (MSRV) and Enzyme Linked Immuno-Sorbent Assay (ELISA) were investigated and compared. For AOAC method two preenrichment medias namely lactose broth and buffer peptone broth, two selective medias namely tetratrionate broth (TTB) and selenite-cystine broth (SC), and two incubation temperatures viz. 35 °C and 42 °C, were evaluated. The result from AOAC method showed that the preenrichment media of lactose broth gave better yield recovery on Salmonella than the buffer peptone water. The selective enrichment media of tetratrionate broth at 42 °C incubation temperature was given better result than TTB at 35 °C and SC at 35 °C The MSRV method is needed 48 hours to detect the Salmonella on shrimp comparison to 96 hours for AOAC. The method of ELISA gave the best results on percentage of recovery, sensitivity and specificity among the three methods. The total time requirement on detection by ELISA was only 28 hours. These methods of detection were tried on contaminated shrimp after washing by chlorinated water (10 ppm) and the results confirmed that the ELISA method has accuracy and specificity agree well with the AOAC method and it demanded very less time for the test. |
