Effect of ganoderma lucidum polysaccharides on growth performance and health of Chinese soft-shelled turtle (Pelodiscus sinensis)

AbstractThe present study aimed to investigate the effects of Ganoderma lucidum polysaccharide (GLP) on the growth and health of the Chinese soft-shelled turtle (Pelodiscus sinensis). The experiment diet (50% crude protein) was supplemented with GLP (50% purity) at 0 (GLP0), 2.5 (GLP25), 5 (GLP50), 7.5 (GLP75) and 10 (GLP100) g/kg feed, respectively. A total of 450 soft-shelled turtles with an initial weight of 4.00 ± 0.20 g were separated into five groups with three replications randomly. The turtles were fed three times per day (8:00, 13:00 and 18:00) with 3% body weight for 8 weeks. At the end of feeding trial, all turtles in each tank were counted and recorded total number and final body weight for growth performance. Three soft-shelled turtles were randomly collected whole blood from the neck for analysing immune parameters, and their whole bodies were stored in a freezer at -20oC for determining nutrient composition. Another three soft-shelled turtles were sampled for whole intestine for indicating microbiota. Besides, the 15 soft-shelled turtles in each tank were continuously evaluated for disease resistance by challenging with Aeromonas hydrophila (2.0×107 CFU/ml). The results showed that the GLP50, GLP75 and GLP100 groups significantly promoted weight gain rate (WGR), specific growth rate (SGR), feed conversion ratio (FCR) and survival rate (SR) compared to the GLP0 group (P<0.05). All levels of GLP supplementation significantly improved the activities of intestinal trypsin and lipase, and the GLP50, GLP75 and GLP100 groups also increased intestinal α-amylase activity compared to the GLP0 group (P<0.05). The serum total superoxide dismutase (T-SOD) activity was significantly enhanced by the GLP50 and GLP75 groups (P<0.05). The serum catalase (CAT) and glutathione peroxidase (GSH-Px) activities were significantly increased, and serum malondialdehyde (MDA) content was significantly decreased by all levels of GLP supplementation (P<0.05). The cumulative morality was significantly decreased by all levels of GLP supplementation after A. hydrophila infection (P<0.05). In conclusion, the GLP supplementation at 7.5 g/kg feed was the recommended level for enhancing growth and health of soft-shelled turtle.
